Epidermal growth factor (EGF) is a potent polypeptide mitogen that initiates its biological activity by interacting with the cell surface. The E G F receptors are both transmembrane proteins with cytosolically exposed tyrosine specific protein kinases which are stimulated by ligand binding. Current data strongly suggests that these kinases are causally linked to many if not all of the biological activities of the EGF receptors. Transmembrane signalling is the result of a composite of ligand-receptor interactions in the endosome (stimulation) and modifications of the primary or quaternary structure of the receptor (activation).
